Samuel was born to Roy Henry Jackson and Elizabeth Harriett in Washington D.C. He was a single child. His father never lived with them and he was an alcohol addict and lost his life to it. He only met him twice in his entire life. Jackson was raised by his mother and maternal grandparents. He initially wanted to be a marine biologist. He was doing his graduation from Morehouse College, Atlanta. There he joined an acting group just for some extra marks and that changed his life. He found his passion and decided to pursue acting as a career.

Jackson and many other students gathered and held the college authorities hostage and put their demands in front of them. Although the college agreed to change Samuel was charged with Unlawful confinement and eventually got convicted. He was suspended for two year from the college. He had been involved in many controversies. He had been a drug addict for a good part of his life. He was admitted to a rehabilitation center during the 1990s. He completed his rehabilitation and gave up drugs.
